Method Transfer - Existing CIEF Methods

Existing CIEF methods from most iCIEF instruments can be transferred to CEInfinite Analytical instrument with no additional method development using the traditional CIEF FC coated capillaries. However, due to the CEInfinite high sensitive detection and the use of the CEInfinite High Performance Imaging CIEF Cartridges (Patent Pending) the resolution can be significantly improved.

Whole Column Time-resolved Imaging Detection Technology - High Resolution Separation

Whole column time-resolved imaging detection (WCTID) capillary electrophoresis (CE) systems have revolutionised protein charge related heterogeneity analysis with capillary isoelectric focusing (CIEF). Unlike the electropherogram in the traditional single point detection CE system display absorbance at a specific point along the separation capillary at different times until completion of the CIEF separation. The CEInfinite iCIEF is your total solution for high resolution CIEF assays. The CEInfinite iCIEF instrument is standard equipped with a high sensitivity CMOS imaging sensor and powerful deep UV LED light source. Using in combination with the CEInfinite High Performance Imaging CIEF Cartridges (Patent Pending) and the CEInfinite AESlytes CAs, which are specially developed and synthesised for high resolution CIEF assays. 

Truly empowers Dynamic Processes Investigation with WCTID

In addition the CEInfinite iCIEF instrument is the only system that truly empowers dynamic processes investigation with WCTID. With the application of a high sensitivity CMOS imaging sensor, scan frequency for the whole separation capillary can be as high as 1000Hz. The increased scan frequency effectively minimises imaging blur that occurs at a lower scan frequency enables dynamic processes, such as

CEInfinite iCIEF High Resolution Fractionation

WCTID CIEF peak fractionation is impossible until the availability of the AES’ Proprietary Cartridge Technology. WCTID CIEF is normally conducted with a short capillary about 5 cm in length. A long enough outlet transfer capillary connected to the separation capillary in WCTID CIEF is necessary to introduce separated proteins into fractionation collection vials. This is impossible to realise with the traditional WCID CIEF cartridges. The resolution achieved in the short separation capillary will be lost during the transfer process when the focused proteins are forced into the transfer capillary.

The CEInfinite Preparative iCIEF instrument provides you an unique, reliable and high resolution protein charged based separation for biologics. Characterisation of individual peaks is essential for comprehending the nature of product isomers and protein degradation pathway. Therefore it is  desirable to perform peak fractionation or direct coupling to mass spectrometer for providing structure characterisation of the separated protein isomer.

Fractionation Purity

The proteins with a difference in pI of 0.2 or greater can be fractionated with greater than 80% purity. Proteins with a difference in pI of 0.1 can be fractionated with greater than 60%.

CEInfinite iCIEF-MS

WCID CIEF direct coupling to MS is impossible until the availability of the AES’ Proprietary Cartridge Technology. WCID CIEF is normally conducted with a short capillary about 5 cm in length. A long enough outlet transfer capillary connected to the separation capillary in WCID CIEF is necessary to introduce separated proteins into MS interface. This is impossible to realise with the conventional WCID CIEF cartridges.

The CEInfinite Preparative iCIEF instrument besides high resolution fractionation enables also direct coupling to MS (patent pending). Protein CIEF separation and mobilisation are monitored with time-resolved imaging of the whole separation capillary, so that high resolution separation of proteins is ensured during focusing and transportation. Complex proteins are separated first according to their isoelectric point in our uniquely built CEInfinite High Performance Imaging CIEF Cartridge and then the separated proteins are forced into the transfer capillary that is connected to the ion source of the mass spectrometer for protein characterisation.

Protein Charged based Separation for Biologics

WCTID CIEF coupled to mass spectrometer (MS) has provided unique, reliable and high resolution protein charged based separation for biologics. Characterisation of individual peaks is essential for comprehending the nature of product isomers and protein degradation pathway. Therefore it is very desirable if WCTID CIEF can be coupled to MS or peak fractionation to provide structure characterisation of the separated protein isomer.


CEInfinite use for MALDI Target Plate

Thanks to the long and small transfer capillary, the CEInfinite Preparative iCIEF instrument allows also to spot the separated protein elution to a MALDI target plate.
