Since plate readers are modular instruments, it is sometimes difficult to choose the optimal plate reader for your needs. Many assays can be performed, many detection modes can be chosen. And that in a full line of plate readers - from relatively simple to high-end. What makes choosing the right plate reader even more challenging is that you are often also constrained by a certain budget.

Affordable, flexible plate reader

One of our commonly sold readers is the SPECTROstar Nano. This is our flexible, dedicated absorbance plate reader for relatively simple assays like Bradford protein detection, ELISA’s, enzyme kinetics and DNA and protein quantification. We also offer a Nanodrop like option to measure DNA, RNA and proteins in down to 2 µl of sample in high throughput. We have a lot of experience in this application field since we also offer the Nanodrop. Sometimes even, fluorescent techniques like Quant-IT are advised because of the limitations of Absorbance based detections. Here, we can offer our fluorescence based plate readers.

High end solutions

What we see in the field is that more and more high-end applications like Fluorescence Polarisation (FP), HTRF, Alpha and Cell based assays are performed on our plate readers. Our CLARIOstar Plus fits these assays perfectly due to its extremely high sensitivity. 

Fluorescence Polarisation

Fluorescence Polarisation (FP) for instance, is a unique and relatively easy technique to determine molecular interactions like protein/protein binding and protein/DNA binding, as well as enzyme activity. Other applications include the analysis of membrane fluidity in liposomes and mitochondrial membranes, and enzymatic assays such as proteolysis, RNA synthesis and ubiquitin conjugation/deconjugation in real time. Fluorescence Polarisation is also used to read our IgG quantification well plates. These 96- or 384- well plates are coated with probes and is based on a homogeneous, single fluorescent label strategy. No sample preparation, additional reagent or washing steps are required.

FP enables direct monitoring of the free/bound ratio of a tracer in solution in real-time. It can be used as end-point detection, equilibrium analysis with very low concentrations (typically down to the picomolar range). FP can also be used for association/ dissociation kinetics.

Technique

Technique in short: Polarised light is sent through the sample in the plate reader. The ratio of polarised/non-polarised light is detected. Small molecules (unbound tracer) will depolarise the polarised light since it is small and has a higher depolarising effect. Bigger molecules do not depolarise polarised light. So when the tracer is bound to the sample, the light stays polarised.

Because FP data is based on the ratio of the bound/unbound tracer, queching or high background fluorescence in the sample have minor effect on the data.
