How does it work?

The latest PCR machine by MBS has been improved in every possible aspect, using MBS Heat transfer technology. These improvements were of such a scale that the inventors named the machine: Next Generation PCR.

1. No ramping times

Traditional PCR the baths are heated and cooled to different temperatures. NextGenPCR®revolutionizes the way in which heat is transferred to the reaction wells using three ‘heating zones’ with a constant temperature corresponding with the desired denaturation, annealing and extension temperatures.

This results in complete removal of the normally required temperature ramping time during which the Peltier element heats or cools the heating block, saving up to 70% of the time required to complete each cycle.

Go from melting to annealing in less than 0.1 second: Additionally, the use of 3 different heating stations creates greater consistency. The instrument has ramp rates of more than 1000 degrees per second. Total PCR reaction time is between 2 and 10 minutes.

2. Ultra fast chemistries

Successful amplification of DNA or RNA sequences in PCR is in part dependent on the quality and speed of the DNA polymerase enzyme used.
Molecular Biology Systems provides several proprietary, ready-to-use PCR master
mixes that can incorporate ≥ 10kb nucleotides per second with excellent accuracy (for other master mixes, have a look at our portfolio).

3. Ultra thin polypropylene

NextGenPCR®technology uses proprietary SBS-format microplates and heat seals that can be compressed by the closing of an upper and a lower heating block in each temperature heating zone. Compression has several beneficial effects to the speed with which the PCR cycle can be completed.

Deformation of the sample well causes microfluidic currents that improve heat transfer from the heating block into the reaction volume and lead to faster annealing of primer oligonucleotide sequences to the target genomic sequence.

End-Point NextGenPCR

The NextGenPCR™ Thermal Cycler is an accurate and energy-efficient thermal cycler designed for ultra-fast PCR. It eliminates temperature ramping times by moving samples between three heating zones, each corresponding to a different temperature. Additionally, it must be used with specially designed microplates made of ultra-thin polypropylene, which allows them to be compressed between the two heating blocks of each heating zone, resulting in instant heat transfer and sample mixing.

PCR time from 92 minutes to 17 minutes

We started using NextGen PCR during the COVID pandemic because we were looking for the fastest way to screen our people for SARS-CoV-2.

In our latest project, we converted a Chlamydia real-time PCR to a NextGen PCR assay and were able to reduce the PCR time from 92 minutes to 17 minutes very quickly.

We introduce many students, the employees of the future, to the equipment. It is very easy to achieve (significant) time savings by using NextGen PCR.

Saving time with PCR: Expediting care to cancer patients

One of the most time-consuming components of next-generation sequencing (NGS) library preparation is polymerase chain reaction (PCR). In our assays, time spent in PCR can sometimes equal or exceed all of the remaining steps of library preparation combined.

Consequently, by drastically reducing the time spent amplifying our genomic regions of interest, we can simultaneously shorten our turnaround time, thereby expediting care to Memorial Healthcare System cancer patients and improving their outcomes.

Essentially eliminating temperature ramping times, Molecular Biology Systems NextGenPCR will allow us to significantly improve turnaround time in all of our PCR-based clinical oncology testing assays, most notably our sequencing panels.

MBS’ NextGenPCR solution for CoV2

Last January 2021, our lab Laboratorio di Servicio implemented and validated the MBS’ NextGenPCR solution for CoV2 screening, in Aruba.

We found this ultra-fast PCR technology easy to use, sensitive, specific and overall, really ultra-fast, both in throughput as well as time to result. Instead of the 300 tests a day, 1000 results per day were produced with a single NG-PCR system.
